Expression of EGFP/RPL10a is driven by Eef1a1, a strong promoter that is widely expressed at high levels in the nervous system and in most tissues.

Detailed description

Cre-mediated excision of a floxed STOP enables tissue-specific expression of EGFP-tagged ribosomal protein that enables transcriptional profiling using TRAP technology. Expression of EGFP/RPL10a is driven by Eef1a1 (eukaryotic translation elongation factor 1 alpha 1), a strong promoter that is widely expressed at high levels in the nervous system and in most tissues. Expression of the endogenous Eef1a1 gene involved in this allele has not been assayed. This strain is particularly useful in detecting proteins expressed at low levels. Homozygotes are not viable.

Progeny of animals crossed with CMV-cre mice demonstrate widespread neuronal colocalization of GFP and Rbfox3 (NeuN, neuron-specific protein), as determined by immunohistochemical (IHC) staining.

Development

A cassette encoding the following was introduced to the first intron of the Eef1a1 gene: loxP-STOP/neomycin-loxP-EGFP/RPL10a (large subunit ribosomal protein L10) cDNA-pA. The mutation was created via homologous recombination in B6(Cg)-Tyr^c-2J/J-derived CY2.4 embryonic stem (ES) cells. Heterozygous mice were backcrossed to C57BL/6J at every generation by the donating laboratory to prevent genetic drift. The Tyr^c-2J albino allele was bred out of the colony. Homozygotes are not viable.

Molecular note: The cDNA encoding a fusion of an EGFP protein with the L10 (large subunit ribosomal protein L10) was inserted into a vector downstream of a floxed 'STOP' cassette and a neomycin selection marker and upstream of a polyadenylation signal. Homologous recombination in ES cells inserted this sequence into the first intron of Eef1a1. Cre-mediated recombination of this allele results in expression of the EGFPL10 protein.
