Description

This bacTRAP transgenic strain, incorporating an EGFP-L10a ribosomal fusion protein targeted to the Ntf3 gene, enables cell labeling and affinity purification of cell-specific polysomal RNAs.

Detailed description

This bacTRAP line incorporates an EGFP-L10a ribosomal fusion protein targeted to the translational start site of the Ntf3 (neurotrophin 3) gene. The fusion protein enables affinity purification of CNS cell-specific polysomal RNAs which can be used for gene expression profiling.

Expression can be detected through immunohistochemistry (IHC) with an antibody against enhanced green fluorescent protein (EGFP). Robust expression of the transgene, recapitulating that of the endogenous gene, is detected in the superficial layer 2/3 of medial cortical areas including medial prefrontal cortex (mPFC), medial orbitofrontal cortex (mOFC), anterior cingulate cortex (ACg), cingulate cortex (Cg), piriform cortex (Pir Cx), septum (Sep), thalamic nuclei such as mediodorsal thalamus (MD), paraventricular thalamus (PV), centromedial thalamus (CM), subregions of hippocampus including dorsal CA3 and dentate gyrus (DG), anterior nucleus of hypothalamus (AHP), anterior pretectal nucleus (APT), and supramamillary nucleus (SuM).

EGFP is directly visible in the medial prefrontal cortex and dentate gyrus. However, immunohistochemistry is recommended to visualize all the EGFP positive cells in the brain.

Development

A shuttle vector was used to introduce sequences encoding an EGFP-L10a ribosomal fusion protein into the translation start site of the Ntf3 gene contained within BAC RP23-79E22. The modified BAC was introduced to FVB embryos. Founders were bred to C57BL/6J mice 4 times by the donating laboratory. This is founder line PS1046. Quantitative PCR suggests 4-5 copies (4 +/- 0.91) of the EGFP transgene are present.
