Description

This CRISPR/Cas9 generated mutant of the Nr4a1 gene carries a knock-out mutation of the Nr4a1 super-enhancer sub-domain E2 (NR4A1se_2) regulatory region (Rr). These mice lack Ly6C^low monocytes while Nr4a1 gene expression is unaltered. This strain may be useful in studies of mononuclear phagocyte system.

Development

Two pairs of Cas9 guide RNA (sgRNA) that target flanking regions of a 1500 bp enhancer roughly centered 4kb upstream of the Nr4a1 gene transcription start site were introduced alongside cas9 nuclease nRNA were introduced into the cytoplasm C57BL/6J-derived fertilized eggs with well recognized pronuclei. The 1500 bp region that is excised in this mutation is the Nr4a1se sub-domain E2, located 4 kb upstream of the Nr4a1 transcription start site. Correctly targeted embryos were transferred to pseudopregnant females. Correctly targeted pups were identified by sequencing and PCR and further bred to C57BL/6J (Stock No. 000664). Heterozygotes were intercrossed to generate homozygotes. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
