Description

PDGFRβ-P2A-CreER^T2 mice express tamoxifen-inducible Cre recombinase under the direction of the murine platelet derived growth factor receptor, beta polypeptide (Pdgfrb) promoter. They are useful for applications requiring tamoxifen-induced deletion of floxed sequences in vascular pericytes.

Detailed description

PDGFRβ-P2A-CreER^T2 mice express cre-ER^T2 directed by the murine platelet derived growth factor receptor, beta polypeptide (Pdgfrb) promoter. Expressed in most mural cells, PDGFRβ is involved in blood vessel development, and the migration and recruitment of pericytes and smooth muscle cells. It also contributes to the remodeling of adipose cells in obese or cold-exposed adult mice. These mice express Cre-ER^T2 protein in pericytes in the vasculature of organs including brain and retina. Restricted to the cytoplasm, cre-ER^T2 can only gain access to the nuclear compartment after exposure to tamoxifen. Homozygotes are viable and fertile. When these mice are bred with mice containing loxP-flanked sequence, tamoxifen-inducible Cre-mediated recombination will result in deletion of the floxed sequences in the Cre-expressing cells of the offspring.

Development

A targeting vector was designed to replace the translation stop codon of the platelet derived growth factor receptor, beta polypeptide (Pdgfrb) gene with a cre-ER^T2 fusion gene (a Cre recombinase fused to a human estrogen receptor ligand binding domain) followed by a frt-flanked neomycin resistance (neo) cassette. A viral P2A oligopeptide, from porcine teschovirus-1, was inserted upstream of the cre-ER^T2 fusion gene to mediate ribosomal skipping. The construct was electroporated into (C57BL/6J x 129S6/SvEvTac)F1-derived KV1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL6/N blastocysts and resulting chimeric males were bred to Tg(ACTFLPe)9205Dym/J females to remove the neo cassette. Offspring were bred together to remove the Flpe-expressing transgene, and PDGFRβ-P2A-CreER^T2 mice were bred to C57BL/6J mice for at least 10 generations. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
