S1P1-R Tg transgenic mice have human CD2 antigen (CD2) promoter/enhancer sequences driving expression of a human sphingosine-1-phosphate receptor 1 (S1PR1) gene. S1PR1 is a G protein-coupled receptor (GPCR) that acts as a regulator of vascular development and function in endothelial cells, and has been shown to play a role in autoimmunity and inflammation. Hemizygotes are viable and fertile. Under control of CD2, S1PR1 is expressed on lymphocytes and acts to modulate the migration, lymphoid tissue distribution, and proliferation of activated lymphocytes. These mice exhibit an increase in chemotaxis of CD4 and CD8 T cells to S1P after anti-CD3 and anti-CD28 activation. S1PR1 signaling in T cells also drives Treg accumulation in tumors, limits CD8+ T cell recruitment and activation, and promotes tumor growth.
