Elf3 (E74-like factor 3) expression is induced by inflammatory factors in a number of cell types. Elevated levels of the protein are found in the cartilage of human osteoarthritis (OA) patients. 

In this transgenic line, a Tet response element (TRE or tetO) was placed upstream of full-length mouse Elf3 cDNA. Crosses with mice expressing tTA or rtTA enable conditional doxycycline-mediated regulation of Elf3 expression in tissues of interest.

To direct expression of Elf3 specifically to adult knee joints, pregnant females crossed with Comp-tTA males, and the resultant pups, may be treated with tetracycline or its analog doxycycline (dox). Postnatal Elf3 overexpression in adult articular cartilage and synovium can be induced through the withdrawl of doxycycline. Increased levels of ELF3 protein in the knees of mice that have undergone destabilization of the medial meniscus (DMM) surgeries significantly increase cartilage degradation at 8 weeks post-surgery.
