Description

Exons 2-8 of the mouse Elf3 gene are flanked by loxP sites in this targeted mutant line. Cre-mediated excision of the floxed region results in a knockout allele. This strain is useful in studies of inflammation and osteoarthritis.

Detailed description

Elf3 (E74-like factor 3) expression is induced by inflammatory factors in a number of cell types. Elevated levels of the protein are found in the cartilage of human osteoarthritis (OA) patients.

Exons 2-8 of the mouse Elf3 gene are flanked by loxP sites in this targeted mutant line. Cre-mediated excision of the floxed region results in a knockout allele. Homozygous floxed mice are viable and fertile, and show no developmental phenotype.

Homozygous knockout mice generated from crosses with Col2a1-cre animals show decreased cartilage degradation in a model of surgically-induced osteoarthritis.

Development

EUCOMM targeting vector HTGRS6002_A_H11 was electroporated into B6(Cg)-Tyr^c-2J/J-derived embryonic stem cells to generate mice carrying an FRT-SA-lacZ-pA-loxP-neomycin-pA-FRT-loxP cassette in intron 1 of Elf3, and a loxP site in intron 8. Resultant chimeric mice were bred to B6(Cg)-Tyr^c-2J/J animals. Subsequent crosses to a mixed B6;SJL background Flp-recombinase-expressing strain (see Stock No. 003800) excised the FRT-flanked neomycin/lacZ cassette, leaving exons 2-8 flanked by loxP sites. This strain was crossed with a mixed B6;129 background Col2a1-cre strain. Although the cre allele has been bred out of the line, the cre strain background contribution remains. This strain was maintained on a mixed genetic background by the donating laboratory. The albino Tyr^c-2J allele is still segregating in the stock.

Molecular note: The L1L2_Bact_P cassette was inserted at position 135258154 of Chromosome 1 (intron 1) upstream of the critical exon(s) (Build GRCm38). The cassette is composed of an FRT site followed by lacZ sequence and a loxP site. This first loxP site is followed by neomycin resistance gene under the control of the human beta-actin promoter, SV40 polyA, a second FRT site and a second loxP site. A third loxP site is inserted downstream of the targeted exon(s) at position 135254725 (intron 8). FLP-mediated recombination excised the FRT-flanked neomycin/lacZ cassette, leaving exons 2-8 flanked by loxP sites.

Molecular note: This mutation has a G-to-T base change at coding nucleotide 230 resulting in an amino acid change from arginine to leucine at residue 77 (p.R77L) which lies in the highly conserved DDRE sequence. Nucleotide 230 is at the alternative 5' splice donor site for exon 1 and this allele does not produce the 1a or 1b subset of tyrosinase transcripts but does produce a significant increase in 1c and 1d transcripts. Western blots of homozygous mutant skin extracts demostrate the nearly complete absence of the broad 76-84 kDa band of glycosylated wild-type tyrosinase. No tyrosinase activity was found in hairbulb extracts from homozygous mice.
