The Pten (phosphatase and tensin homolog) gene is a dual specificity protein phosphatase and encodes a tumor suppressor that functions through negative regulation of the AKT/PKB signaling pathway. Mutations in PTEN are associated with multiple primary malignant tumors. Embryos homozygous for the knock-out allele have a lethal phenotype; exhibiting abnormal patterning in cephalic and caudal regions and defective placentation at E8.5. Death occurs by E9.5 due to defective chorio-allantoic development. Mice heterozygous for the mPTEN3-5 allele have a high incidence of T cell lymphomas associated with loss of heterozygosity of the wild-type allele. Tumor formation is accelerated by gamma-irradiation and is associated with elevated phosphorylation of AKT/PKB. These mice may be useful for studies of cancer progression.
