Description

Tg-MBP1-EGFP transgenic mice express a human myelin basic protein (MBP) fragment/EGFP fusion protein in neural cells. Tg-MBP1-EGFP may be used to assess the effect of myelin basic protein and its derived fragments on fibrillary amyloid pathology and behavioral performance.

Detailed description

Tg-MBP1-EGFP transgenic mice have the mouse Thy1 sequences driving expression of a human myelin basic protein (MBP) fragment (containing amino acids 1-64) EGFP fusion protein to neural cells. Homozygotes are viable, fertile. MBP expression is modest and does not alter levels of endogenous mouse protein. When combined with mice carrying Tg(APPSwFlLon,PSEN1*M146L*L286V)6799Vas (5X-FAD) (MMRRC Stock No. 034848), progeny exhibit an improvement in spatial learning memory and a reduction in soluble Aβ and fibrillar amyloid plaque size. Tg-MBP1-EGFP may be used to assess the effect of myelin basic protein and its’ derived fragments on fibrillary amyloid pathology and behavioral performance.

Development

A human myelin basic protein (MBP) N-terminal fragment containing amino acids 1-64 and fused to EGFP at its C-terminus was subcloned into exon 2 of the mouse Thy1 expression cassette. The Thy1 cassette was altered to remove exon 3 and its flanking introns, limiting expression to neural cells. The vector was introduced to C57BL/6N embryos. Founder line C was selected to generate homozygotes. This strain is maintained on a C57BL/6N background.
