Description

Transgenic Cacna1gHIGH males overexpress Cacna1g and are useful for studying pathways and development of epilepsy and ataxia.

Detailed description

Hemizygous males and homozygous females carrying the Cacna1gHIGH transgene are viable and fertile, with overexpression of the entire calcium channel α 1g (Cacna1g) gene directed by the endogenous Cacna1g promoter/enhancer regions on the BAC transgene. Transcript expression of Cacna1g in hemizygous males is approximately threefold that of wild-type males. These mice do not exhibit overt neurological abnormalities associated with complex absence epilepsy. Cacna1gHIGH transgenic mice are useful for studying pathways and development of epilepsy and ataxia.

Development

The bacterial artificial chromosome (BAC) RP23-65I14 containing the entire calcium channel α 1g (Cacna1g) gene (also containing ATP-binding cassette transporter gene, Abcc3, nonchannel protein, Ankrd40, gene and the 3' end of Spata20) was injected into C57BL/6 fertilized oocytes. Mice from the founder line designated Cacna1gHIGH (or B6.1GH) were established and the transgene was found to have inserted on the X chromosome. Mice were maintained on the C57BL/6J background.
