Description

This Ki67^cre/ERT2 knock-in strain expresses tamoxifen inducible Cre recombinase in actively dividing cells.

Detailed description

The targeted Mki67 gene encodes a nuclear protein that is a marker for proliferating cells. These mice carry an inducible form of Cre recombinase (cre/ERT2) knock-in allele of the Mki67 gene. When crossed to a reporter strain, tamoxifen-inducible cre/ERT2 activity can be detected in the spleen, thymus, brain, hair follicles, epidermis, bone marrow, eye, liver, pancreas, stomach, kidney, heart, lung, small intestine and colon. Most actively dividing cells in the adult brain exhibit tamoxifen-inducible cre/ERT2 activity. Mice that are homozygous for the targeted mutation are viable and fertile. The Donating Investigator reports that presence or absence of the neo selection cassette does not alter the phenotype.

Development

A targeting vector containing ires2-creERT2 and a SV40 polyadenylation site followed by a FRT site flanked PGKneo cassette was used to introduce cre/ERT2 at the C-terminus of the Mki67 gene. The construct was electroporated into 129P2/OlaHsd-derived IB10 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. The resulting chimeric animals were crossed to FLP recombinase expressing mice on an unknown genetic background, and then backcrossed to C57BL/6J for 3 generations. Mice that no longer contained the neo selection cassette (determined by PCR testing) were used to generate homozygotes. The mice were then backcrossed to C57BL/6J for 3 generations. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
