Detailed description

RIAM^flox mice possess loxP sites flanking exons 3-4 of the amyloid beta (A4) precursor protein-binding, family B, member 1 interacting protein (Apbb1ip) gene. Also known as proline-rich protein 48 (Prp48), RIAM is a member of the MRL family of adaptor proteins that includes lamellipodin. It is essential for efficient adhesion of lymphocytes to ICAM-1 and VCAM-1 and for proper trafficking of lymphocytes to secondary lymphoid organs. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exons 3-4 of the RIAM gene deleted in cre-expressing tissues.

When bred to B6.C-Tg(CMV-cre)1Cgn/J mice (Stock No. 006054) with widespread Cre Recombinase expression, resulting RIAM^-/- KO mice have normal T- and B-cell development, but peripheral lymph nodes are depleted of both B and T cells. Recirculating B cells are diminished in the bone barrow. The spleen RIAM^-/- KO mice is hypercellular and IgM levels are significantly decreased.

Development

The RIAM^flox targeting vector was designed to insert a loxP site upstream of exon 3, and a frt-flanked neomycin resistance (neo) cassette, followed by a second loxP site, downstream of exon 4 of the amyloid beta (A4) precursor protein-binding, family B, member 1 interacting protein (Apbb1ip) gene. The construct was electroporated into (C57BL/6 x 129S4/SvJae)F1-derived V6.5 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6NTac blastocysts and resulting chimeric males were bred with C57BL/6J females. Offspring were bred with B6;SJL-Tg(ACTFLPe)9205Dym/J mice (Stock No. 003800) to delete the neo cassette. Resulting RIAM^flox mice were crossed to remove the Flp-expressing allele, and were subsequently maintained on a mixed background. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
