Description

Exon 2 of the C1ql3 is flanked by loxP sites upstream of an mVenus fluorescent reporter in this conditional mutant strain. Cre-mediated excision of the floxed region results in a knockout allele and prevents mVenus expression.

Detailed description

C1ql3 (C1q-like 3) promotes excitatory synapse formation and maintenance in vitro and in vivo and is required for implicit association memories of emotionally salient stimuli (e.g. fear memory).

Exon 2 of mouse C1ql3 is flanked by loxP sites upstream of an mVenus fluorescent reporter in this conditional mutant strain. C1QL3/mVenus, strongly enriched in presynaptic terminals, is broadly expressed throughout the brain (including all cortical areas tested), but only in discrete subsets of neurons, including excitatory projection neurons of the basolateral amygdala and the insular and perirhinal cortices. mVenus co-localizes with the pan-neuronal markers MAP2 and Hu, and with the excitatory neuronal marker CaMKIIα, but not with the inhibitory neuronal marker GAD67. Cre-mediated excision of the floxed region results in a C1ql3 knockout allele and prevents mVenus expression.

Constitutive ablation of C1ql3 via crosses with a CMV-cre recombinase-expressing strain (see Stock No. 006054), produces animals that show diverse behavioral changes, including hyperactivity, impairments in motor learning, and a deficit in association learning of rewarding and aversive stimuli.

Homozygous floxed reporter mice are viable and fertile, born at the predicted Mendelian ratio, and exhibit normal body weight.

Development

A loxP-Frt-neomycin-Frt cassette was introduced to intron 1 and loxP-IRES-mVenus cassette was placed in intron 2 (the 3' UTR) via homologous recombination in an unspecified 129S6 x C57BL/6J F1-derived embryonic stem (ES) cell line. Resultant mice were crossed with a C57BL/6 background Flp-expressing strain to excise the neomycin cassette, leaving exon 2 flanked by loxP sites. This strain was backcrossed to C57BL/6J for 3 generations by the donating laboratory.
