Description

This Pla2g2a variant allele (knock-out) strain is useful in studies of inflammation and rheumatoid arthritis.

Detailed description

Phospholipase A2, group IIA (platelets, synovial fluid), encoded by the Pla2g2a gene, regulates phospholipid metabolism in biomembranes and is involved in eicosanoid biosynthesis, inflammation, rheumatoid arthritis, sepsis, psoriasis, pancreatitis and cancer. Deletion of the Pla2g2a gene has been found to enhance intestinal polyp number in Apc^min mice on a C57BL/6J genetic background (PMID: 8242739). These mice carry a spontaneous variant allele of the Pla2g2a gene, Mom1-s (modifier of Min1), in which a T insertion at position 208 (position 166 from the ATG) in exon 3 results in a frameshift which produces a stop codon 13 amino acids downstream in exon 4. No gene product (mRNA) is detected by Northern blot or RT-PCR analysis of small intestine from mutant mice. Mice that are homozygous for the mutation on this congenic BALB/cJ background, are viable and fertile. When challenged with K/BxN serum treatment, homozygous mice develop a less severe autoantibody-driven arthritic response compared to controls.

Development

The Pla2g2a^Mom1-s insertion mutation is found in 129, C57BL/6J and BTBR inbred strains. First described in C57BL/6J, the Pla2g2a^Mom1-s is a T insertion at position 208 (position 166 from the ATG) in exon 3 that results in a frameshift which produces a stop codon 13 amino acids downstream in exon 4. Dr. Barbara Balestrieri (Brigham and Women's Hospital) obtained the Pla2g2a^Mom1-s allele from 129X1/SvJ (Stock No. 000691) and backcrossed the mice to BALB/cJ (Stock No. 000651) for 11 generations.

Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize BALB/cJ oocytes (Stock No. 000651).

A mutation in the Apc gene of the mouse causes multiple intestinal adenomas in the C57BL/6J strain on which it arose. In crosses with other strains, the number of tumors is reduced. A locus controlling about 50% of genetic variation in tumor number has been identified as Pla2g2a.
