5-HT1A^-/- mice have a neo cassette inserted downstream of the third transmembrane domain of the 5-hydroxytryptamine (serotonin) receptor 1A (Htr1a) gene. These mice may be useful for studying serotonin receptor signaling.

Detailed description

5-HT1A^-/- mice have a neo cassette inserted downstream of the third transmembrane domain of the 5-hydroxytryptamine (serotonin) receptor 1A (Htr1a) gene, abolishing gene function. 5-HT1A is expressed in the central nervous system, in the cerebral cortex, hippocampus, septum, amygdala, and raphe nucleus in high densities. Binding of serotonin to this receptor is associated with mood, depression, anxiety, learning, and behavior. Homozygotes are viable and fertile and lack functional serotonin receptor 1A. They exhibit increased levels of anxiety-related behaviors. They also show increased levels of serotonin and dopamine.

On this BALB/cJ background, mice are also useful when studying light-induced retinopathy.

Development

A targeting vector was designed to insert a neomycin resistance (neo) cassette downstream of the third transmembrane domain of the 5-hydroxytryptamine (serotonin) receptor 1A (Htr1a) gene. The construct was electroporated into 129S1/Sv-Oca2⁺ Tyr⁺ Kitl⁺-derived W9.5 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6J blastocysts, and the resulting chimeric males were bred to 129/Sv females and subsequently to BALB/cJ mice for at least 9 generations. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize BALB/cJ oocytes (Stock No. 000651).
