Description

These 5●OR1A1Cβ transgenic “MouSensor” mice express an IRES-tau-mCherry tagged human OR1A1, olfactory receptor family 1 subfamily A member 1, gene, and exhibit enhanced odor-detection of the OR1A1 ligand. These mice may be useful in studies related to human olfaction.

Detailed description

These 5●OR1A1Cβ transgenic mice express the an IRES-tau-mCherry tagged human OR1A1, olfactory receptor family 1 subfamily A member 1, gene in approximately 13% of the olfactory sensory neurons. Transgene expression is detected in the main olfactory epithelium. Hemizygous mice exhibit a 2-log decrease in the detection threshold for the high affinity OR1A1 ligand, (-)-Carvone, in a two-bottle behavioral avoidance task. Hemizgyotes have approximately 18 copies of the transgene. Homozygotes are viable and fertile.

Development

The nx21-OR-IRES-Reporter MouSensors transgenic vector is designed to allow insertion of a 21-bp singular gene enhancer, odorant receptor coding sequence, and IRES-Reporter cassette. This 5●OR1A1Cβ transgene was generated from the nx21-OR-IRES-Reporter MouSensors transgenic vector and contains 5 repeats of the 21bp enhancer sequence (from the olfactory receptor H-region enhancer sequence), and an IRES-tau-mCherry tagged human OR1A1, olfactory receptor family 1 subfamily A member 1, gene. The 5●OR1A1Cβ transgene was injected into fertilized CBA X C57BL/6 mouse eggs. Founder line D736-A8 was subsequently established. The mice were then crossed to 129 wildtype mice. Hemizgyotes have approximately 18 copies of the transgene. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
