Detailed description

This strain has exon 5 deleted, which disrupts the reading frame and eliminates both the N-terminal EF-hand domain and the C-terminal mitochondrial membrane-bound transporter domain. These Slc25a13 null mice have no overt phenotypic abnormalities. Homozygotes are viable and fertile.

Development

Working in C57BL/6J-derived ES cells, exon 5 was replaced with a loxP flanked neomycin selection cassette in the laboratory of Dr. Kenneth Johnson at The Jackson Laboratory. The founder was bred to C57BL/6J generating the C57BL/6J-Slc25a13^tm2Kjn/Kjn strain. This was bred to B6N.Cg-Tg(Sox2-cre)1Amc/J and backcrossed just once to C57BL/6J to remove the transgene then maintained by sibling breeding. This excised the neomycin selection cassette and generated this strain. Sperm was cryopreserved from homozygous males at generation N1F5.
