Description

These UPK II-Cre transgenic mice have the mouse uroplakin 2 (Upk2) gene promoter driving Cre recombinase expression in the bladder urothelium. These mice may be useful for Cre-lox studies of urothelium function and cancers of the bladder.

Detailed description

Uroplakin 2, encoded by the Upk2 gene, is part of the urothelial apical plaque, or asymmetric unit membrane of the bladder epithelium, and can be detected in the peripheral blood of patients with transitional cell carcinomas. These transgenic mice express Cre recombinase under the control of the mouse uroplakin 2 (Upk2) gene promoter. When crossed with with Rosa-R26R-YFP reporter mice, Cre recombinase activity is detected in the urothelium of the bladder and ureter.

During backcrossing, the Y chromosome may not have been fixed to the C57BL/6J genetic background Hemizygotes are viable and fertile. The Donating Investigator has not attempted to make the strain homozygous. Homozygous viability/fertility has not been tested (August, 2016).

Development

A transgenic construct containing Cre recombinase (cre) under the control of the 3.6 kb mouse Upk2, uroplakin 2, gene promoter, was injected into fertilized C57BL/6 mouse eggs. Founder line 001 was subsequently established. The transgenic mice were crossed to another mutant line on the B6;129 background. The mice were then backcrossed to C57BL/6 for 2 generations. During backcrossing, the Y chromosome may not have been fixed to the C57BL/6 genetic background. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
