These Slm1 floxed mutant mice possess loxP sites flanking exon 2 of the Khdrbs2 gene. This strain may be useful for generating conditional mutations in applications related to the regulation of alternative splicing.

The RNA-binding protein encoded by the Khdrbs2 gene is found in a subset of interneurons of the brain (hippocampal glutamatergic dentate granule cells, a subset of hippocampal
cholecystokinin–calbindin double-positive inhibitory interneurons, cerebellar Purkinje cells) and is involved in alternative splicing regulation.
These Slm1 floxed mice possess loxP sites on either side of exon 2 of the targeted gene. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exon 2 deleted in the cre-expressing tissues.
Removal of the floxed sequence creates a null allele.
When bred to a strain with Cre recombinase expression in Purkinje cells, this mutant mouse strain may be useful in studies of neuronal alternative splicing.
A targeting vector containing a FRT site flanked PGK-Neo selection cassette was utilized in the construction of this mutant. This selection cassette was inserted downstream of exon 2 of the targeted gene, and another loxP site was inserted upstream of exon 2. This construct was electroporated into unspecified 129SvEvTac derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts.
The resulting chimeric animals were crossed to ROSA-26 Flpe mice expressing FLPe recombinase from the endogenous Gt(ROSA)26Sor locus. Mice that retained the loxP site flanked exon 2 were then bred to C57BL/6 mice to remove the Gt(ROSA)26Sor FLPe recombinase knock-in alelle. Heterozygotes were crossed to generate homozygotes. The mice were then backcrossed to C57BL/6 for 9 generations.
Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
