Itpkc-/- mice lack 1.1 kb of the first exon of the inositol 1,4,5-trisphosphate 3-kinase C (Itpkc) gene. The mice have an overtly normal phenotype and may be useful for studying T cell development.

Itpkc-/- have a neo cassette replacing 1.1kb of the 3’ end of exon 1 of the inositol 1,4,5-trisphosphate 3-kinase C (Itpkc) gene, abolishing gene expression. ITPKC catalyzes the phosphorylation of inositol 1,4,5-trisphosphate to 1,3,4,5-tetrakisphosphate, which is essential for T lymphocyte development. ITPKC negatively regulates T-cell activation. A mutation in human ITPKC has been associated with susceptibility to Kawasaki disease, a severe and acute pediatric systemic vasculitis. Homozygous mice are viable and fertile. No differences are evident in multiciliated tracheal epithelial cells and sperm cells compared to wild type mice.
Itpkc-/- mice exhibit normal thymocyte development.
T cell development and calcium response following stimulation are also unremarkable. Mice are overtly normal and do not appear to recapitulate Kawasaki disease traits.
A targeting vector was designed by Dr. Stephane Schurmans (Universite de Liege) to replace 1.1 kb of the 3’ end of exon 1, and a short fragment of the sequence following intron 1, of the inositol 1,4,5-trisphosphate 3-kinase C (Itpkc) gene with a neomycin resistance (neo) cassette in reverse orientation to the gene. The construct was electroporated into (129X1/SvJ x 129S1/Sv)F1-Kitl+-derived R1 embryonic stem (ES) cells. Correctly targeted ES cells were aggregated with CD1 morulae. The resulting chimeric males were bred to C57BL/6NCrl females. Itpkc-/- mice were maintained on a mixed background. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304).
