Zfp191flox mice possess loxP sites flanking exons 2-3 of the zinc finger protein 24 (Zfp24) gene. ZFP191 is a transcription factor that may be required for the conversion of oligodendrocytes from the premyelinating
to the myelinating state. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exons 2-3 deleted in cre-expressing tissues.
A targeting vector was designed to insert a single loxP site upstream of exon 2, and a loxP-flanked neomycin resistance (neo) cassette downstream of exon 3 of the zinc finger protein 24 (Zfp24) gene. The neo cassette was also flanked by frt sites. The construct was electroporated into C57BL/6-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts. Resulting chimeric mice were bred with mice expressing Cre recombinase under direction of the protamine promoter, on B6 background, to remove the neo cassette. Resulting offspring were crossed to remove the cre-expressing transgene. These Zfp191flox mice were maintained on a C57BL/6 background. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
