Impdh2loxP mice possess loxP sites flanking exons 2-9 of the inosine 5'-phosphate dehydrogenase 2 (Impdh2) gene. IMPDH2 is an enzyme that catalyzes the conversion of inosine 5-phosphate (IMP) to xanthosine 5-phosphate (XMP), the first committed and rate-limiting step in the de novo synthesis of guanine nucleotides during cell growth. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exons 2-9 deleted in the cre-expressing tissues.
For example, when bred to Wnt1-Cre transgenic mice (Stock No. 003829), expressing Cre recombinase in in the midbrain and developing neural tube, homozygous mice live to term but do not survive after birth. They display defects in multiple neural crest derivatives including highly penetrant intestinal aganglionosis, near absence of the cranio facial skeleton, and cardiac outflow tract and great vessel malformations.
A targeting vector was designed to insert a frt-flanked neomycin resistance (neo) cassette and a loxP site upstream of exon 2, and a second loxP site was inserted downstream of exon 9 of the inosine 5'-phosphate dehydrogenase 2 (Impdh2) gene. The construct was electroporated into 129P2/OlaHsd-derived E14TG2a embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and resulting chimeric males were bred with C57BL/6J females. Subsequently, offspring were bred with B6;SJL-Tg(ACTFLPe)9205Dym/J mice (Stock No. 003800) to delete the neo cassette, and progeny were crossed to remove the Flp-expressing transgene. Resulting Impdh2loxP mice were bred to C57BL/6J mice (Stock No. 000664) for many generations before being bred to a Swiss albino strain. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes.
