These Pax8YFP^tg transgenic mice express yellow fluorescent protein in developing kidney nephron tubules.

Detailed description

The transcription factor encoded by the Pax8 (paired box 8) gene is important in kidney and thyroid follicular cell development. These Pax8YFP^tg transgenic mice express yellow fluorescent protein under the control of the Pax8 promoter. Yellow Fluorescent Protein immunofluorescence is observed in nephron tubules formed during metanephric kidney development. YFP expression is detected in other tissues where Pax8 expression is observed, though these have not been fully characterized. The Donating Investigator reports that there is no difference between the phenotype exhibited by homozygotes compared to hemizygotes.

Homozygotes are viable and fertile.

As the mice are characterized, we will modify the strain description and add phenotype data.

Development

The 166 kb C57BL/6J mouse bacterial artificial chromosome (BAC) from the RPCI23 library containing the entire Pax8 locus (and other genes) was modified by targeting a yellow fluorescent protein (YFP) coding sequence into the ATG start site of Pax8. This inserted a YFP protein coding sequence, SV40 polyA signal, and a FRT-flanked Kanamycin resistance gene cassette. No other genes on the BAC were modified or deleted. Correctly targeted clones were identified and the selection cassette was removed using FLP recombination. This modified BAC clone (vector pBACe3.6) was linearized and microinjected into CD-1 zygotes. The BAC clone backbone vector (pBACe3.6), which contains the chloramphenicol resistance (CmR) gene, was not removed prior to microinjection and thus is retained in the transgenic animals. Transgenic founders were bred to C57BL/6 mice to establish the Pax8YFP-tg colony, and were possibly outcrossed to Swiss Webster prior to strain donation.

Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
