Description

TNF-α^glo mice harbor a loxP-flanked EGFP cassette that prevents transcription of a CAG promoter-driven Tnf cDNA. TNF-α is only expressed following Cre-mediated recombination. This strain may be useful when studying inflammatory pain.

Detailed description

TNF-α^glo transgenic mice hemizygous for this conditional allele are viable and fertile. Mice have a CMV-IE enhancer/chicken beta-actin/rabbit beta-globin hybrid promoter (CAG) driving expression of a loxP-flanked enhanced green fluorescent protein (EGFP) and a mouse tumor necrosis factor (Tnf) cDNA. TNF-α is a cytokine that promotes inflammation by recruitment of leukocytes, inducing vasodilation, and stimulating the production of proinflammatory cytokines. Elevated levels of TNF-α have been associated with infections and autoimmune disorders. TNF-α expression is determined by which tissues express Cre recombinase. When bred to mice that express Cre recombinase, the resulting offspring will have the loxP-flanked EGFP cassette deleted in the cre-expressing tissue(s).

When bred with B6N.FVB-Tg(Dmp1-cre)1Jqfe/BwdJ transgenic mice (Stock No. 023047), expressing Cre Recombinase in odontoblasts and osteocytes, resulting offspring overexpress TNF-α in the tooth pulp and bone. Mice exhibit inflammation within the tooth pulp, including inflammatory infiltrates, disruption of the odontoblast layer, and dilated blood vessels. They also show alveolar bone loss around the tooth root.

Development

The TNF-α^glo transgene was designed by Dr. Ashok Kulkarni (National Institute of Dental and Craniofacial Research, National Institutes of Health) with (from 5' to 3') a CMV-IE enhancer/chicken beta-actin/rabbit beta-globin hybrid promoter (CAG), a loxP-flanked fusion protein containing an enhanced green fluorescent protein sequence (EGFP), and a polyadenylation (pA) signal, a mouse tumor necrosis factor (Tnf) cDNA followed by a two pA signals. The transgene was microinjected into fertilized FVB/N oocytes and founder mice were bred to FVB/N mice. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize FVB/NJ oocytes (Stock No. 001800).
