Calp mutant mice possess loxP sites flanking exons 4-9 of the Capns1 or calpain, small subunit 1 gene. Calpains are calcium-dependent cysteine proteinases involved in multiple cellular functions; in particular they have been implicated in neurodegenerative diseases. Heterozygous mice are viable and fertile, however, homozygous embryos exhibit lethality prior to E10.5. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exons 4-9 deleted in the cre-expressing tissues. This allele results in embryonic lethality in the homozygous state.
For example, when bred to mice carrying a pan-neuronal CRE3 transgene, mice exhibit impaired performance in the Morris Water Maze (MWM). This strain may be useful for studying protein function in neuronal injury.
A loxP-flanked neomycin resistance cassette is inserted upstream of exon 4 followed by a loxP site downstream of exon 9. The construct was electroporated into unspecified embryonic stem (ES) cells. Correctly targeted ES cells were injected into recipient blastocysts and the resulting chimeric animals were crossed to FVB/N mice for at least 13 generations. Subsequently, mice were backcrossed to C57BL/6 for more than 10 generations. At some point, this strain was combined with a C57BL/6 congenic carrying Tg(Mylpf-cre)3Kraj, Birc5tm1Wnt, and Ctnnb1tm2Kem. These alleles were later bred out to establish this line. Upon arrival at The Jackson Laboratory, mice were bred to C57BL/6J inbred mice for at least one generation to establish the colony.

While maintaining a live colony, these mice are bred as heterozygotes, specifically the donating investigator recommends heterozygous female x wild-type male. This allele results in embryonic lethality in the homozygous state.
