This bacTRAP line expresses EGFP-RPL10a from the mouse Slc6a4 promoter. Ribosome-bound mRNAs from serotonergic neurons can be purified using EGFP as an affinity tag.

Detailed description

Slc6a4 (solute carrier family 6 (neurotransmitter transporter, serotonin), member 4) encodes a protein that transports serotonin from the synaptic cleft to the presynaptic neuron and is a common target of many antidepressant medications.

This bacTRAP line expresses EGFP-RPL10a from the mouse Slc6a4 promoter. Ribosome-bound mRNAs from serotonergic neurons can be purified using EGFP as an affinity tag. Immunohistochemistry with anti-GFP antibodies is reported to demonstrate robust expression in a pattern consistent with expression in all serotonergic neurons. Expression was also found in the dorsal part of the caudal spinal trigeminal nucleus. Neurotransmitter phenotype was confirmed using confocal immunofluorescence with antibodies to GFP and serotonin. In all cells, GFP signal is seen only in the cytoplasm and nucleolus, consistent with ribosomal incorporation of the fusion protein.

Development

EGFP-RPL10a was inserted at the translation start site of the Slc6a4 gene encoded by BAC RP24-335M24. The modified BAC was injected into FVB eggs. Resultant founder mice (founder line JD60) were crossed with C57BL/6J animals for 3 or 4 generations by the donating lab.
