In response to insulin the Solute Carrier Family 2 (Facilitated Glucose Transporter), Member 4, also known as the Insulin-Responsive Glucose Transporter Type 4 (GLUT4) is rapidly translocated from intracellular storage sites to the muscle cell surface, allowing uptake of glucose. Mutations in the human SLC2A4 gene have been linked to noninsulin-dependent diabetes mellitus.
These transgenic mice express the rat Slc2a4, solute carrier family 2 (facilitated glucose transporter), member 4, sequence with a 14 residue sequence of the myc-epitope (GLUT4myc), under the control of the mouse Ckm, creatine kinase, muscle, gene promoter. Transgene expression is detected predominantly in the I-band region of skeletal muscle fibers, with colocalization with desmin in the Z-disk, and is approximately 3 fold higher than endogenous protein levels in skeletal muscle. In the presence of insulin, there is a 2.1 fold increase in myc-epitope at the surface of dissociated, nonpermeabilized muscle fiber segments or of a preparation of giant membrane/sarcolemmal vesicles. Muscle fiber contraction also increases translocation of the transporter to the cell surface (2.3 fold), as well as glucose uptake (2.6 fold). Combined insulin and contraction stimuli increases cell surface translocation by 3.9 fold and glucose uptake by 5.1 fold in giant sarcolemmal vesicles.
In glucose tolerance tests transgenic mice have lower blood glucose levels compared to wildtype controls.
During backcrossing, the Y chromosome may not have been fixed to the C57BL/6 genetic background. Mice that are homozygous for the transgene are viable and fertile. 
The Donating Investigator reports that homozygotes over 4 months of age are not efficient breeders.

These MCK-GLUT4myc transgenic mice express a myc-tagged rat Slc2a4 in skeletal muscle tissue, allowing for quantitative analysis of SLC2A4 glucose transporter translocation to the cell surface, and co-localization studies. This strain may be useful in studies of insulin action and resistance, and glucose uptake.
