Downregulation of the miR-143/145 cluster (also called Mirc32, microRNA cluster 32, including Mir143 and Mir145a) has been reported in colon cancer and other epithelial tumors. Overexpression of these miRNAs inhibits tumorigenesis, suggesting they function as cell-autonomous epithelial tumor suppressors.
Mirc32 is flanked by loxP sites in this miR-143/145flox/+ targeted mutant strain. MiR-143/145 microRNA is detected at normal levels in homozygous floxed animals. Cre-mediated excision of the cluster results in a null allele.
A germline deletion allele (miR+/-; see Stock No. 028504) is also available. Germline deletion of these miRNAs results in no overt developmental defects and the targeted allele is transmitted at expected Mendelian ratios. Animals lacking Mirc32 show normal intestinal development, but epithelial regeneration after injury is dramatically impaired.
A loxP site was inserted upstream of Mir143 and an FRT-flanked neomycin cassette and loxP site were inserted downstream of Mir145a via homologous recombination in 129-derived embryonic stem (ES) cells. The neomycin cassette was excised through crosses with an ACTB-Flp strain, leaving the miR sequences flanked by loxP sites. This strain was backcrossed to C57BL/6J for 15 generations by the donating lab.
