Detailed description

IL1R^loxP/loxP floxed mice possess loxP sites flanking exons 3-4 of the interleukin 1 receptor, type I (Il1r1) gene. IL1R1 encodes the cytokine receptor for IL-1, a proinflammatory cytokine that drives immune and inflammatory responses after tissue injury or infection. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exons 3-4 deleted in the cre-expressing tissues.

Development

A targeting vector was designed by Dr Randy Blakely (Vanderbilt University Medical School) to insert a loxP site upstream of exon 3, containing the translation initiation sequence, and a second loxP site downstream of exon 4 of the interleukin 1 receptor, type I (Il1r1) gene. A frt-flanked neomycin resistance (neo) cassette was placed downstream of the 3’ loxP site. The construct was electroporated into 129-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and resulting chimeric mice were bred with Flp transgenic mice on a C57BL/6 background to delete the neo cassette. Progeny were crossed to remove the Flp-expressing transgene, and resulting IL1R^loxP/loxP mice were bred to C57BL/6J mice for 10 generations. Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
