Transmembrane channel-like gene family 1 (Tmc1) is essential for mechanoelectrical transduction (MET) in inner ear sensory hair cells.
These TMC1-mCherry BAC transgenic mice express the mouse Tmc1 gene and an mCherry fluorescent reporter under the control of the endogenous Tmc1 promoter. Expression of the transgene was confirmed in the presumed site of mechanolelectrical transduction at stereocilia tips. Confocal fluorescence images of inner hair cells from TMC1-mCherry transgenic mice at postnatal day 8 (P8) show the transgenic protein localizes predominantly to the tips of the shorter rows of stereocilia.
The Tmc1 stop codon (c. 2272-2274) of mouse bacterial artificial chromosome (BAC) MSMG01-526L10 was replaced with cDNA encoding mCherry. No other genes are located on this BAC. The modified BAC was purified and injected into (C57BL/6 x SJL) F2 mouse embryos. Founder mice carrying the transgene were intercrossed with other targeted and transgenic alleles (see Stock Nos. 028517, 019146, and/or 019147). The TMC1-mCherry transgene was subsequently bred away from the other mutations. Line 2, carrying 7 copies of the transgene was backcrossed from a mixed background (C57BL/6, SJL, 129) to C57BL/6 for more than 5 generations by the donating lab.

Molecular Note | The transgene was created by targeted insertion of mCherry protein coding sequence into the bacterial artificial chromosome (BAC) MSMG01-526L10. The mCherry sequence was integrated at the stop codon of Tmc1, the only gene in the BAC. The integration site was not investigated. This C-terminal fusion is expressed in the sensory hair cells of the inner ear. Confocal fluorescence images of inner hair cells from transgenic mice at postnatal day 8 (P8) show the transgenic protein localizes predominantly to the tips of the shorter rows of stereocilia. Line 2 carries 7 copies of the transgene. |
