Description

CBG transgenic mice have the surfactant associated protein C (Sftpc) promoter/enhancer sequences on a BAC transgene driving expression of EFGP. These mice may be useful for identification, tracking, and isolation of alveolar epithelial type II cells.

Detailed description

CBG transgenic mice have the mouse surfactant associated protein C (Sftpc) promoter/enhancer sequences driving expression of EGFP. Homozygotes are viable, fertile. Under control of Sftpc, EGFP labels all alveolar epithelial type II cells in the lung. Cells can be isolated by FACS and fluorescence can be detected for at least 14 days in culture.

Development

Bacterial artificial chromosome (BAC) library (CHORI) was used to obtain a BAC (#RP23–247J9) containing the entire mouse surfactant associated protein C (Sftpc) gene. This BAC was modified to insert an internal ribosome entry site (IRES) fused to an enhanced green fluorescent protein (EGFP) sequence into the 3’ UTR of the Sftpc gene. This CBG (SPC-BAC-EGFP) transgene was microinjected into fertilized C57BL/6xDBA2 F1 oocytes and hemizygous founder mice, containing between two and four copies of the transgenic BAC per haploid genome, were bred to C57BL/6NCrl mice to establish a colony. CBG transgenic mice were bred to C57BL/6NCrl mice for at least 30 generations. Upon arrival at The Jackson Laboratory, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6NJ oocytes (Stock No. 005304).

Of note, the BAC used in the development of this strain contained other genes including: Epb4.9, Rai16, Nudt18, Hr, Reep4, Lgi3, BMP1, Phyhip, and one tRNAala gene. It is unknown if these genes are still active.
