This allele was generated by inserting a conditional by inversion (COIN) cassette containing a rabbit beta-globin polyA site, EGFP and inverted lox site variants (lox71/lox66). A plasmid was synthesized containing 1 kb homology arms and two silent mutations (c.24C>T and c.27G>T). Double-stranded plasmid DNA, Cas9 mRNA, and sgRNA were introduced into C57BL/6NJ zygotes via pronuclear injection. The founder was backcrossed to C57BL/6NJ and sperm was cryopreserved from heterozygous males at generation N3.
