Description

These Cd23-cre transgenic mice express cre under the control of the Fcer2a promoter and may be useful for generating conditional knockouts in B cells specific for studying B cell development and differentiation.

Detailed description

These Cd23-cre transgenic mice express cre under the control of the Fcer2a promoter. Cre recombinase expression is detected in immature, transitional and late B cells in the bone marrow and spleen. Mice that are hemizygous for the transgene are viable and fertile. The Donating Investigator has not attempted to make the strain homozygous. During backcrossing, the Y chromosome may not have been fixed to the C57BL/6J genetic background.

Development

The mouse bacterial artificial chromosome (BAC) (RP24-321J16) containing the entire Fc receptor, IgE, low affinity II, alpha polypeptide (Fcer2a or Cd23) gene, was modified by inserting a cre recombinase gene, with the cre linked to a truncated human CD5 indicator gene by an IRES, and a SV40 polyadenylation site into exon 2. The cre recombinase gene is under the control of the Fcer2a promoter. DNA from this modified BAC clone was used as the source for the transgene, which was microinjected into the pronucleus of (C57BL/6 x CBA)F1 fertilized eggs. Founder line 5 was subsequently established. The mice were backcrossed to the C57BL/6J background for more than 20 generations. During backcrossing, the Y chromosome may not have been fixed to the C57BL/6J genetic background.

Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
