The PAX8 transcription factor is important in kidney and thyroid follicular cell development. Mice that are heterozygous for the targeted mutation are viable and fertile. Homozygotes exhibit a postnatal lethal phenotype. Cre activity is similar to the wildtype gene expression pattern and is detected in the inner ear by E16.5; the brain beginning at E9.5; the mesonephros starting at approximately E9.5; geniculate ganglions at E10.5; thyroid gland, vestibulocochlear nerve, epididymis, and adrenal gland at E15.5. During backcrossing, the Y chromosome may not have been fixed to the C57BL/6J genetic background.
A targeting vector containing the cre recombinase gene, a FRT site flanked NEO cassette and SV40 polyadenylation site was used to disrupt exon 3 of the targeted gene. The construct was electroporated into 129P2/OlaHsd derived E14.1 embryonic stem (ES) cells.
Correctly targeted ES cells were injected into C57BL/6 blastocysts.
The resulting chimeric male animals were bred to C57BL/6 females to generate Pax8neo/+ mice.
The mice were backcrossed to C3H/He for 4 generations.
The mice were then crossed to the FLPe recombinase-expressing deleter strain on the C57BL/6 genetic background to remove the FRT flanked NEO cassette, producing the Pax8cre. The mice were then backcrossed to C57BL/6J for more than 20 generations. During backcrossing, the Y chromosome may not have been fixed to the C57BL/6J genetic background.
Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).

Molecular Note | Crossing Pax8tm1(cre)Mbu with mice expressing FLP recombinase in the germline created this allele in which the frt-flanked neo downstream of the inserted Cre was excised, allowing Cre to be expressed. Expression of cre recombinase was detected in the known Pax8 expression domains; in addition, expression was detected in the adrenal gland, and in the facial, vestibulocochlear and cuneate nerves. |
When maintaining a live colony, heterozygous mice may be bred together, to wildtype siblings, or to C57BL/6J inbred mice (Stock No. 000664). Homozygotes are not viable.
