Description

These floxed mutant mice possess loxP sites flanking exons 17 through 19 of the Tcf3 gene, and a FRT site flanked GFP-NEO cassette downstream of exon 19. This strain may be useful for generating conditional mutations in applications related to hematopoiesis, and as a reporter for E2A (Tcf3) expression.

Detailed description

These Tcf3^fl (Tcfe2a^fl) mice possess loxP sites on either side of exons 17 and 19, as well as a FRT-site flanked GFP-NEO cassette downstream of exon 19, of the targeted gene. Mice that are homozygous for this allele are viable and fertile. When these mutant mice are bred to mice that express Cre recombinase, resulting Tcf3^gfp offspring will have exons 17 and 19 deleted in the cre-expressing tissues and expression of an E2A-GFP fusion protein. Removal of the floxed sequence creates a null allele. Sequential Cre and FLP recombination, results in the deletion of the floxed exons 17 through 19 and the FRT site flanked GFP-NEO cassette.

When bred to a strain with Cre recombinase expression in hematopoietic cells and progenitor hematopoietic cells (see Stock No. 008610 for example), this mutant mouse strain may be useful in studies of B lymphogenesis and labelling and quantification of B lymphocytes by GFP expression.

Development

A targeting vector containing FRT site flanked GFP-neo cassette was utilized in the construction of this mutant. This selection cassette with a 5’ loxP site and 6 SV40 Poly-adenylation regions was inserted downstream of exon 19 of the targeted gene, and another loxP site was inserted upstream of exon 17. This construct was electroporated into 129P2/OlaHsd-Hprt^b-m3 derived HM-1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts. The resulting chimeric animals were tested for germline transmission. The resulting chimeric animals were crossed to C57BL/6J or 129/Sv mice, and then backcrossed to C57BL/6J for 10 generations.

Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize C57BL/6J oocytes (Stock No. 000664).
