The Pax6flox allele has loxP sites flanking exon 4-6 of the paired box 6 gene. Removal of the floxed sequence creates a null allele. These mice may be useful in studying the role of PAX6 in the eye, nose, pancreas and central nervous system.

The Pax6flox allele has loxP sites flanking the initiation codon in exon 4 and the paired domain (exon 6). Pax6, a member of the Pax family, encodes a homeobox and paired domain-containing protein that functions as a transcription factor and is expressed in the eye, nose, pancreas and central nervous system. Mice homozygous for the floxed allele are viable and fertile with no reported abnormalities.
When bred to mice that express Cre recombinase, the resulting offspring may be useful in generating a tissue-specific Pax6 knockout.
A targeting vector was designed to insert a loxP site in exon 4 upstream of the first ATG and a loxP site followed by an FRT-flanked neomycin cassette between exons 6 and 7 of the Pax6 (paired box 6) gene. The construct was electroporated into (129X1/SvJ x 129S1/Sv)F1-derived R1 embryonic stem (ES) cells. Correctly targeted ES cells were aggregated with NMRI-derived morula. Chimeric mice were bred to NMRI mice. Offspring were bred with ACTB:FLPe (Bmpr1bTg(ACTB-FLP)4917Dym) transgenic mice to delete the neomycin cassette. The resulting progeny were backcrossed to C57BL/6N mice in excess of 10 generations. Upon arrival at The Jackson Laboratory, mice were bred to C57BL/6NJ inbred mice for at least one generation to establish the colony.

|Molecular Note||Insertion of loxP sequences flanking the exons encoding the amino terminus of the protein, including the initiator methionine and most of the paired domain. A frt-flanked neomycin cassette in intron 6 was removed via Flp-mediated recomination to create the final allele. These insertions had no effect on the normal function of the allele.|
While maintaining a live colony, these mice are bred as homozygotes.
