The Tmem151b^nmf389 mutation results in fewer dendritic processes from the CA pyramidal cells of the hippocampus and a decreased electroconvulsive threshold.

Detailed description

TMEM151B (transmembrane protein 151B) is highly conserved in vertebrates and predicted to be a 561 amino acid protein with 2 transmembrane domains and a molecular mass of 62kDa that is expressed in the hippocampal pyramidal cell layer and dentate gyrus, the Purkinje cell layer and internal granule cell layer, and has low expression in the cerebral cortex. This 42 nucleotide deletion results in a recessive in-frame loss of 14 amino acids (YHERVNTHVAEAEF), but transcript expression levels were not found to be altered. Although not significantly sensitive to seizures induced by kainic acid or pentylenetetrazole, both heterozygotes and homozygotes have a lowered threshold for electroshock-induced seizures and spontaneous seizures have been observed in mice over 3 months of age subsequent to handling. There are fewer dendritic processes from the CA pyramidal cells of the hippocampus, which may underlie the seizure phenotype. This lowered seizure threshold is also found on the coisogenic C57BL/6J background.

Development

The nmf389 phenotype was found in the offspring of an ENU (N-ethyl-N-nitrosourea)-mutagenized C57BL/6J male and was maintained on the C57BL/6J background for many generations. It was subsequently backcrossed onto C57BL/6NJ and sperm was cryopreserved in 2015 at generation N10F3. This 42 nucleotide deletion in exon 3 of the mouse Tmem151b gene results in a recessive in-frame loss of 14 amino acids (YHERVNTHVAEAEF), but transcript expression levels were not found to be altered.
