Smarca4 (also known as Brg) is fused to Dendra2, a green-to-red, irreversibly photoswitchable monomeric fluorescent protein.

Detailed description

Smarc4 (SWI/SNF related, matrix associated, actin dependent regulator of chromatin, subfamily a, member 4; also called Brg) is the ATPase subunit of the chromatin-remodeling Brg-associated factor (BAF) complex. It plays an essential role in early T cell development.

Dendra2 is a green-to-red, irreversibly photoswitchable monomeric fluorescent protein derived from octocoral Dendronephthya sp. Its fusion with Smarca4 in this knock-in strain enables nuclear fluorescence which can undergo photoswitching in cells/tissues expressing the protein. Expression was confirmed by the donating lab via Western blot using anti-Brg (SMARCA4) and anti-Dendra2 antibodies. Cells show nuclear fluorescence which can undergo photoswitching.

Development

The Dendra2 sequence, along with a floxed neomycin cassette, was inserted into the C-terminus of the Smarca4 gene via homologous recombination in 129-derived embryonic stem cells. The targeting construct was derived from BAC bMQ-242m22 (source: BioScience). The neomycin cassette was later excised. This strain was maintained on a mixed C57BL/6-129 genetic background by the donating laboratory.
