Detailed description

Dyrk1a (dual-specificity tyrosine-(Y)-phosphorylation regulated kinase 1a) catalyzes autophosphorylation of serine/threonine and tyrosine residues. Knockouts of the mouse gene interfere with embryonic growth and viability.

In this targeted mutant strain, the exon encoding Dyrk1a's kinase active site has been flanked by loxP sites. Mice homozygous for the floxed allele are viable, fertile, and overtly normal. Cre-mediated excision of the floxed region creates a knockout allele.

Development

LoxP-flanked neomycin resistance cassettes were introduced both 5' and 3' of the exon that encodes the kinase active site (the fifth coding exon). The 5' neomycin cassette was excised with cre recombinase during vector construction in E. coli. The vector was introduced to 129S6/SvEvTac-derived TC1 embryonic stem (ES) cells, and the mutation was created through homologous recombination. Resultant chimeric mice were crossed with EIIa-Cre animals (see Stock No. 003724) to excise the 3' floxed neomycin cassette, leaving the kinase active site exon flanked by loxP sites. This line was backcrossed to C57BL/6J for more than 5 generations by the donating lab.
