Description

These Npas1-tdTom mice express icre and tdTomato in most (approximately 88%) of Npas1-expressing neurons in the brain. They are suitable for use in studies related to the external globus pallidus (GPe) nucleus in the basal ganglia.

Detailed description

These transgenic mice express codon-optimized icre and tdTomato under the direction of the mouse Npas1, neuronal PAS domain protein 1, gene promoter. Transgenic expression is detected in neurons of the external globus pallidus (GPe) nucleus in the basal ganglia, cortex, hippocampus, basal magnocellular complex, internal globus pallidus, zona incerta, substantia nigra pars reticulata, and superior colliculus. Some neurons in the dorsal striatum and thalamus also express the transgene. Expression of the transgene is similar to the endogenous expression pattern of Npas1. In founder line 1, approximately 88% of Npas1 expressing neurons are also tdTomato-positive. Hemizygotes are viable and fertile. Homozygous viability/fertility has not been tested (November, 2016).

Development

A mouse bacterial artificial chromosome (BAC) containing the entire Npas1 gene, was modified by inserting Cre-2A (cleavable fusion peptide sequence)-tdTomato and polyadenylation sequence from bovine growth hormone downstream of the ATG translation initiation codon, and removing Npas1 coding sequence. A transgenic insert made from this modified BAC was microinjected into the pronucleus of C57BL/6 fertilized eggs. The resulting Npas1-tdTom transgenic mice from founder line 1 were subsequently maintained on the C57BL/6J background. Embryos were cryopreserved. Upon arrival, to establish our live colony, embryos were cryorecovered.
