Sox7 (SRY (sex determining region Y)-box 7) encodes a transcription factor involved with the regulation of embryonic development that is expressed in vascular endothelial cells.
The coding region of the mouse Sox7 gene is flanked by loxP sites in this conditional mutant strain. Floxed animals demonstrate no overt phenotype. Cre-mediated excision of the floxed region results in a knockout allele that expresses cyan fluorescent protein (CFP) and hemagglutinin tags. Germline recombination leads to mid-embryonic lethality due to defects in cardiovascular/hematopoietic development.
LoxP sites were inserted (1) within the first exon 5’ of the initiator methionine codon, and (2) 3’ of an SV40 polyadenylation signal that was inserted immediately 3’ of the 3’ UTR. The following DNA segments were added immediately downstream of the 3’ loxP site: (1) the coding region for CFP with a 3×HA tag, and (2) a phosphoglycerate kinase (PGK)-Neo cassette flanked by frt sites (FNF), where a third loxP site was inserted immediately 5’ of the second frt site. The targeting construct was electroporated into 129-derived ES cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts. Resulting chimeric males were crossed to C57BL/6 mice and then germline Flp mice (see Stock No. 003800) to excise the frt-PGK-Neo-frt (FNF) cassette. This strain was backcrossed to C57BL/6J for 3 generations by the donating lab.
