Tph2f/f floxed mice may be useful for studying the role of serotonin in the maintenance of serotonin (5-HT) neurons and their effect on psychiatric and developmental disorders.

Tph2f/f floxed mice possess loxP sites flanking exon 5 of the tryptophan hydroxylase 2 (Tph2) gene. TPH2 is an enzyme essential for the synthesis serotonin in the brain, which is required for neurogenesis, differentiation, and survival of central serotonin (5-HT) neurons. Serotonin is involved in numerous central nervous activities including the regulation of mood, appetite, sleep, memory and learning. Compromised serotonin neuronal phenotype may contribute to abnormal physiologic function and the onset of psychiatric disorders such as depression, anxiety and schizophrenia. In peripheral tissues, serotonin has a role in the maintenance of vascular tone and gut motility. Mice that are homozygous for this allele are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exon 5 deleted in the cre-expressing tissues.
A targeting vector was designed to insert a loxP site upstream of exon 5, with a frt-flanked neomycin resistance (neo) cassette followed by a second loxP site downstream of exon 5 of the tryptophan hydroxylase 2 (Tph2) gene. The construct was electroporated into 129S7/SvEvBrd-Hprt+-derived AB1 embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts and resulting chimeric males were bred with wildtype females. The resulting Tph2f/f mice were maintained on a mixed background. Upon arrival, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.
