Gpr177Fx floxed mice possess loxP sites flanking exon 3 of the wntless homolog (Wls) gene. Mice that are homozygous for this allele are viable and fertile. The Wls gene encodes a chaperon protein with a role in regulation of Wnt protein sorting and secretion, anterior-posterior body axis development, proper formation of the skin, skeletal development and disease, tooth morphogenesis, craniofacial development and deformity, mammary gland morphogenesis and Wnt-induced tumorigenesis. When these mutant mice are bred to mice that express Cre recombinase, resulting offspring will have exon 3 deleted in cre-expressing tissues.
For example, when crossed to B6.Cg-Tg(Wnt1-cre)11Rth Tg(Wnt1-GAL4)11Rth/J mice expressing Cre recombinase in the neural tube and midbrain (Stock No. 009107), resulting mice are embryonic lethal and exhibit mid/hindbrain and craniofacial defects, including cleft palate.
A targeting vector was designed to insert a loxP site upstream of exon 3 and a loxP-flanked neomycin resistance (neo) cassette downstream of exon 3 of the wntless homolog (Wls) gene. The construct was electroporated into 129S6/SvEvTac-derived CSL3 embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and resulting chimeric mice were bred together. Progeny were bred to B6.FVB-Tg(EIIa-cre)C5379Lmgd/J transgenic mice (Stock No. 003724) to delete the neo cassette. Resulting offspring contained multiple gene rearrangments; intact floxed-exon 3, intact floxed-neo cassette, or excision of both exon 3 and the neo cassette. Gpr177Fx mice, containing only the floxed-exon 3, were bred with C57BL/6 mice. Upon arrival, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.
