These Nms-iCre mice express codon-improved Cre recombinase in a subset of Nms-expressing neurons in the suprachiasmatic nucleus and may be useful in studying circadian rhythms and behavior.

The Nms gene encodes the neuromedin S protein which is expressed in the suprachiasmatic nucleus (SCN) of the hypothalamus and plays a role in regulation of circadian period. Mice hemizygous for the Nms-iCre transgene are viable, fertile, and normal in size. These mice carry a transgene containing a codon-improved Cre recombinase gene in the 3’ UTR of neuromedin S (Nms) sequence in the transgenic construct. Cre recombinase expression is restricted to a subset of Nms expressing neurons in the SCN. The Donating Investigator has not attempted to make the strain homozygous.
The mouse bacterial artificial chromosome (BAC) RP23-101P2 containing the entire Nms gene, was modified by inserting an IRES2-iCre cassette into the 3’ UTR of Nms. DNA from a modified BAC clone was used as the source for the Nms-iCre transgene, which was linearized and then microinjected into the pronucleus of C57BL/6N oocytes. The resulting transgenic mice from founder line 20 were subsequently crossed to C57BL/6J prior to arrival at The Jackson Laboratory. Upon arrival, the mice were crossed to C57BL/6J (Stock No. 000664) at least once to establish the colony.
A 48 SNP (single nucleotide polymorphism) panel analysis, with 43 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. One of the 43 markers, on Chromosome 12, was segregating.
