Description

These ARR₂Pb-Ron (Line B) transgenic mice overexpress Mst1r in the prostate epithelium and develop prostate adenocarcinoma and microinvasive prostate intraepithelial neoplasia.

Detailed description

The macrophage stimulating 1 receptor, also known as RON (Recepteur d'Origine Nantais) kinase, regulates cell survival, migration and differentiation, playing an important role in epithelial and stromal tumorigenesis as well as tumor microenvironment. It is overexpressed in human breast cancer, many carcinomas and prostate cancer. These ARR₂Pb-Ron (Line B) transgenic mice selectively overexpress Mst1r in the prostate epithelium. The transgene consists of a wildtype mouse Mst1r minigene under the control of the ARR2Pb prostate-specific promoter (consisting of two androgen response elements from the rat probasin gene promoter arranged sequentially). Overexpression of Mst1r gene product is detected by Western blot analysis and quantitative RT-PCR of prostates from transgenic mice. Measurable over typical histopathology (atypical hyperplasia, prostate intraepithelial neoplasia, prostate intraepithelial neoplasia with invasion, and adenocarcinoma) of the prostate is observed in 75% of transgenic mice at 1 year of age and in 100% of transgenic mice after 450 days of age. Atypical prostate hyperplasia progesses to adenocarcinoma in 20% of transgenic mice. Mice that are homozygous for the transgene are viable and fertile.

Development

A transgenic construct containing a wildtype mouse Mst1r minigene under the control of the ARR₂Pb prostate-specific promoter consisting of two androgen response elements from the rat probasin gene promoter arranged sequentially, was injected into fertilized FVB/N mouse eggs. The full-length wildtype mRon minigene consists of genomic DNA sequence of the first 3 exons and introns, and cDNA sequence encoding exons 4 though 19. Founder line B was subsequently established. Upon arrival at The Jackson Laboratory, the mice were crossed to FVB/NJ (Stock No. 001800) at least once to establish the colony.
