Description

The moZP2^Trunc strain carries a Zp2 KO allele and a mouse ZP2 BAC transgene in which exons 2-5 and part of 6 (a.a. 51-149) have been removed resulting in female sterility. This strain may be useful for studying the role of the ZP2 N-terminus in human gamete recognition.

Detailed description

The moZP2^Trunc strain carries a Zp2 KO allele and a mouse ZP2 BAC transgene in which exons 2-5 and part of 6 (a.a. 51-149) have been removed. This region of the N-terminal includes the gamete recognition domain necessary for sperm binding. The zona pellucida from these truncated ZP2 female mice is thinner than normal and cannot bind sperm. As a result female moZP2^Trunc mice are sterile. This strain may be useful for studying the role of the ZP2 N-terminus in human gamete recognition.

Development

The full length sequence of the mouse Zp2 gene in BAC RP23-6513 was modified by removing DNA encoding amino acids 51-149 (bp 59 in exon 2 to bp 27 in exon 6) at the N-terminus of gene. The linearized BAC was injected into FVB/N oocytes to generate transgenic animals. Mice from the founder line were crossed to mice carrying the Zp2^tm1Dean allele on a mixed background. Upon arrival, mice were bred to FVB/N for at least 1 generation to establish the colony.
