Description

The hu/moZP2^Rescue strain is homozygous for the Zp2 KO allele and a mouse Zp2 BAC transgene in which exons 2-5 and part of exon 6 are replaced with the corresponding human DNA from exons 2-5. This strain may be useful for studying the role of the ZP2 N-terminus in human gamete recognition.

Detailed description

The hu/moZP2^Rescue strain is homozygous for the Zp2-null allele and a mouse Zp2 BAC transgene in which exons 2-5 and part of exon 6 are replaced with the corresponding human DNA from exons 2-5. This region of the N-terminal includes the gamete recognition domain necessary for sperm binding. The zona pellucida appears normal in ovarian sections from female mice. Human sperm binds to the zona pellucida from these mice but not to the zona from the reciprocal mo/huZp2^Rescue strain. This strain may be useful for studying the role of the ZP2 N-terminus in human gamete recognition.

Development

The full length sequence of the mouse Zp2 gene in BAC RP23-6513 was modified by replacing the corresponding human exons encoding amino acids 41-68 (bp 26 in exon 2 to bp 21 in exon 6) for the mouse amino acids 37-165 (bp 59 in exon 2 to bp 6 in exon 5) at the N-terminus of gene. The linearized BAC was injected into FVB/N oocytes to generate transgenic animals. Mice from the founder line were crossed to mice carrying the Zp2^tm1Dean allele on a mixed background. Upon arrival, mice were bred to FVB/N for at least 1 generation to establish the colony.
