ShcWT transgenic mice have human eukaryotic translation elongation factor 1 alpha 1 (EEF1A1) promoter/enhancer elements driving expression of a flag-tagged wildtype human src homology 2 domain-containing transforming protein C1 (SHC1) cDNA fused to an IRES-EGFP fusion protein. SHC1 cDNA is preceded by a loxP-flanked SV40 polyadenylation signal allowing transcriptional control of the SHC1 protein. SHC1 acts downstream of many different receptors including growth factor receptors, integrins, TCR, BCR, and the preTCR. Interaction of SHC1 with these receptors leads to phosphorylation of three SHC1 tyrosine residues, Y239, Y240, and Y317 in the central proline-rich domain. Phosphorylation of these residues allows recruitment and binding of SH2-domain containing proteins that lead to downstream Ras-MAPK pathway signaling. Positive and negative selection of thymocytes at the double postitive stage requires signaling through the mature TCR and molecules in the Ras-MAPK pathway. Homozygous mice are viable and fertile. After cre-excision of the floxed-STOP sequence, these mice express flag-tagged human SCH1. For unknown reasons, GFP fluorescence is not detectable, therefore, tracking of cells expressing the ShcFFF mutant via GFP fluorescence as a surrogate marker is not possible with this strain. 
 
These mice serve as a wildtype control for B6.129-Tg(EEF1A1-SHC1*)1Ravi/J (Stock No. <a href="https://www.jax.org/strain/025880">025880</a>) mice, which express cre-inducible human SHC1 containing mutations in all three tyrosine residues, resulting in a dominant negative transgene.

The ShcWT transgene in this strain is a cre-inducible form of wildtype human SHC1 and may be useful in studies examining SHC1 requirements during development, T cell maturation, and tumorigenesis.
