mSYD1A^flox mice may be useful for studying synaptogenic signaling and vesicle docking.

Detailed description

mSYD1A^flox mice contain loxP-sites flanking exons 2-4 of the synapse defective 1, Rho GTPase, homolog 1 (Syde1) gene. mSYD1A protein forms a complex with presynaptic receptor tyrosine phosphatases and controls tethering of synaptic vesicles at synapses. The gene lies on Chr 10 in a region that is mutated in human disorders with autistic features. The inactivation of mSYD1 affects the behavior of mice which could bridge the missing link in the fundamental processes of autistic disorders. Homozygous mice are viable and fertile. When bred to mice that express tissue-specific Cre recombinase, resulting offspring will have exons 2-4 deleted in the cre-expressing tissues. Synapses in mSYD1A knock-out hippocampus exhibit a severe reduction in morphologically docked vesicles and reduced synaptic transmission.

Development

C57BL/6N-A^tm1Brd-derived JM8A3.N1 embryonic stem (ES) cells containing a targeted mutation of the synapse defective 1, Rho GTPase, homolog 1 (Syde1) gene were obtained from The European Conditional Mouse Mutagenesis Program (EUCOMM). The Syde1 gene has, from 5’ to 3’, a frt site, an En2 splice acceptor cassette containing a β-galactosidase sequence, and loxP-flanked neomycin cassette, containing a frt site, inserted downstream of exon 1. A third loxP-site is inserted downstream of exon 4. Correctly targeted ES cells were injected into C57BL/6J blastocysts and resulting chimeric mice from clone HEPD0665-7-C10 were bred to ROSA26-Flp transgenic mice to remove the frt-flanked β-galactosidase and neo cassettes. Offspring were crossed to remove the Flp-expressing transgene and mSYD1A^flox mice were bre to C57BL/6J mice for at least 6 generations. Upon arrival, mice were bred to C57BL/6J inbred mice (Stock No. 000664) for at least one generation to establish the colony.
