Description

These GUS_8.4GFP mice express red-shifted Green Fluorescent Protein, under the control of the mouse Gnat3 gene promoter, in taste receptor cells.

Detailed description

The transducin-like G-protein subunit encoded by the Gnat3 gene, also called alpha-gustducin, is responsible for taste signal transduction (bitter, sweet and umami/glutamate). These GUS_8.4GFP transgenic mice express red-shifted Green Fluorescent Protein under the direction of the mouse Gnat3 gene promoter. Taste receptor cells from circumvallate papillae and retinal bipolar cells (type 7 ON cone bipolar cells and weakly in rod bipolar cells) fluoresce green. The Donating Investigator reports that there is no difference in phenotype between hemizygous (carrier) and homozygous transgenic mice. Mice that are homozygous for the transgene are viable and fertile.

Development

A transgenic construct containing red-shifted Green Fluorescent Protein (GFP) under the control of the 8.4 kb mouse Gnat3, guanine nucleotide binding protein, alpha transducing 3, (alpha-gustducin), gene promoter, was injected into fertilized B6CBAF2/J zygotes. Founder line 1 was subsequently established. The donating investigator reported that the mice were then backcrossed to 129S1 for more than 9 generations (see SNP note below). Upon arrival, sperm was cryopreserved. To establish our live colony, an aliquot of frozen sperm was used to fertilize 129S1/SvImJ oocytes (Stock No. 002448).

A 32 SNP (single nucleotide polymorphism) panel analysis, with 27 markers covering all 19 chromosomes and the X chromosome, as well as 5 markers that distinguish between the C57BL/6J and C57BL/6N substrains, was performed on the rederived living colony at The Jackson Laboratory Repository. Three of the 27 markers throughout the genome were segregating, suggesting an incomplete backcross. Also, 1 of 5 markers that determine C57BL/6J from C57BL/6N were found to be segregating. These data suggest the mice sent to The Jackson Laboratory Repository contained C57BL/6J and C57BL/6N in their genetic background.
